Proline transport by rabbit ciliary body-iris in vitro.

چکیده
The rabbit ciliary body-iris preparation in vitro shows concentrative uptake of C-proline which is resistant to anoxia and to cyanide; inhibited by ouabain; and abolished in the absence of external sodium or by cold (0° C). It has a Q,o (27 to 37° C.) of 1.7. Uptake is decreased by preloading with unlabeled proline. Exchange diffusion of the accelerative type is demonstrable during efflux of >C-proline. Inhibitory effects of other neutral amino acids in this tissue differ from effects in rabbit choroid plexus and optic nerve. In the ciliary bodyiris preparation the neutral amino acids do not divide themselves into a high-affinity group, the imino acids, and a group with lesser affinity. Neither arginine nor aspartic acid inhibits the uptake of >C-proline. Saturable proline uptake is nearly completely inhibited by 50 mM N-methylalanine. Thus, proline is concentrated in the ciliary body-iris preparation by a mobile carrier system largely of the Ehrlich cell A type. Alanine uptake is only partly inhibited by N-methylalanine, and the residual saturable uptake is only partly sodium dependent. The ciliary body-iris preparation, therefore, transports alanine by three carrier systems like those described in the Ehrlich cell as the A, the ASC, and the L mediations. The functional role of these carrier systems is discussed.

The accumulation of alpha aminoisobutyric acid by rabbit ciliary body-iris preparations.
H.-labeled alpha aminoisobutyric acid (AIB) was accumulated against a concentration gradient by rabbit ciliary body-iris preparations in vitro. The accumulation ratio tissue per medium was greater in the ciliary processes than in the iris. The process was saturable and energy and temperature dependent. Accumulation ratio was depressed by other amino acids, metabolic inhibitors, and when Na+ or ...
متن کاملIncreased concentration of glucocorticoid receptors in rabbit iris--ciliary body compared to rabbit liver.
Dexamethasone binding to both rabbit iris-ciliary body and rabbit liver glucocorticoid receptors was compared by means of a batch assay in which the binding components were adsorbed to hydroxylapatite. The results showed that homogenates of both the rabbit iris-ciliary body and the rabbit liver contained a single class of high-affinity receptors. Binding affinity was virtually identical for the...
متن کاملAge variation of iodopyracet and iodide accumulation by the rabbit ciliary body-iris preparation.
In adult rabbits there is an active transport of ascorbic acid into the aqueous humor: the concentration of ascorbic acid in the anterior chamber is about 20 times that in plasma. In very young animals, however, the anterior chamber level differs little from the plasma level. Recently it has been demonstrated that iodopyracet (Diodrast) and iodide are actively transported out of the eye in vivo...
متن کاملIntracameral anesthesia: in vitro iris and corneal uptake and washout of 1% lidocaine hydrochloride.
OBJECTIVES To characterize the uptake, washout, and metabolism of lidocaine hydrochloride in the iris/ciliary body and cornea. METHODS Iris/ciliary body uptake of lidocaine hydrochloride was measured by incubating human and rabbit irides in radiolabeled carbon 14-1% lidocaine hydrochloride for 2 to 60 minutes. Washout was determined by incubating the iris in 14C-1% lidocaine hydrochloride for...
متن کاملOcular disposition of aminozolamide in the rabbit eye.
We have previously determined that 6-amino-2-benzothiazolesulfonamide (aminozolamide) significantly lowers IOP in rabbits and, more importantly, in ocular hypertensive human subjects. Results from in vitro experiments established that both the inhibitory activity of aminozolamide against carbonic anhydrase B as well as the penetration rate across excised rabbit corneas were equivalent to ethoxz...
